TNF is produced upon activation of monocytes and DCs by microbial products like lipopolysaccharide (LPS) of Pam₃Cys, which act by binding to toll like receptors (TLRs). When cells are exposed to these compounds repeatedly, then the TNF production is decreased, i.e. the cells have become tolerant. The molecular mechanism of tolerance to date has been studied only in monocytes and macrophages. For LPS stimulation it was shown early on that the CD14 co-receptor was not down but rather up-regulated in tolerant cells [1]. Also, signal transduction still occurred with mobilisation of p50/p65 of NF-κB, but there was at the same time an increase in NF-κB p50-homodimers which bind to the promoter and displace the p50/p65 heterodimer. Since p50 cannot transactivate this will lead to blockade of TNF gene expression [1,2]. In addition to this p50-homodimer mechanism, blockade in LPS tolerant monocytes can also occur through interruption of the signalling cascade at the level of IRAK-1 in that this adaptor protein is proteolytically degraded [3,4]. For monocytes tolerant to the TLR-ligand Pam₃Cys this mechanism also applied and there was a strong and complete ablation of IRAK-1 [5].

How tough it is to figure out that these crooks are lying their eyeballs out about "Lyme Disease?"

There are TWO DIFFERENT DISEASES: Steere's HLA-Associated Hypersensitivity-TooManyAntibodies-Disease, and what everyone else has- their own special New Great Imitators. Everyone with Chronic Lyme usually has three primary New Great Imitators - usually the MS version of Lyme (from B cell mutations from, typically, Esptein Barr, caused by Pam3Cys gumming up the normal-immunity-works) plus "Chronic Fatigue Syndrome" plus a Delirium - and at least one other secondary New Great Imitator, like thyroid disease, or cancer, or a pseudolymphoma... and these TWO DIFFERENT DISEASES are: 1) one that causes no problems (which is the one that tests Steere's Dearborn positive and needs no expensive treatment), and 2) what everyone else has, the multiple Imitators, which is clearly disabling.

Now who would be interested in the non-treatment of illnesses?

Now clearly Yale knew how to detect Lyme and were looking for MOST CASES of LYME, regardless of one's genetic background, and clearly they knew how to validate a method, and clearly they knew that most people with Lyme had the one reliable band, 41 or flagellin, and clearly they went about making it SPECIFIC to BURGDORFERI or "Lyme Spirochetes"...

I guess people are not understanding this: Each antibody band is as accurate for diagnosis as its assigned percent specificity. The Yale flagellin method is 100% or nearly 100% SPECIFIC, and as most people with Lyme have this band 41, 17/18 % of the cases is as Accurate as we will ever get in an antibody test for Lyme.

I understand Pam Weintraub refutes the FDA and their Bioanalytics Division Rules on the Validation of an Analytical Method.
I hope she has the sense to take her arguments up with the FDA and stops attacking Lyme victims for repeating what those rules
are, and also demonstrating what Yale claims to have developed as a validated method for detecting Lyme Borreliosis- a method that is SPECIFIC for Lyme Borrelia and detects 17/18 cases:

CHAPTER 3, STEERE/DEARBORN - How Steere in Europe falsified the CDC's diagnostic standard (blood testing criteria for a "case" of "Lyme Disease") in 1994 to A) deny neurologic disease diagnosis (needs expensive IV treatment) and B) set up the racketeering monopoly on tick borne diseases (test kits and vaccines) around OspA or the LYMErix vaccine and the associated genetic markers of outcomes (this has bioweapons significance). No one looks into the overall problem of "test kits."

CHAPTER 7, BRAIN PERMANENT The infection is permanent and could be seen as a Trojan Horse due to its syphilis-like latency.
